Biological Background: RGS2 Function and Localization
- RGS2, also known as G0S8 (G0/G1 switch regulatory protein 8) and cell growth-inhibiting gene 31 protein, is a regulator of G protein signaling that accelerates GTPase activity of G protein alpha subunits, driving them to inactive GDP-bound state, thereby inhibiting signal transduction. Related references: PMID:11063746 PMID:19478087
- It negatively regulates the angiotensin-activated signaling pathway, contributing to blood pressure control and vascular smooth muscle constriction/relaxation. Related references: PMID:28784619
- RGS2 also inhibits mRNA translation by binding to EIF2B5 and blocking its activity. Related references: PMID:19736320
- Subcellularly, RGS2 localizes to the cell membrane, cytoplasm, nucleus, nucleolus, and mitochondrion.
- It is expressed in acute myelogenous leukemia (AML) and acute lymphoblastic leukemia (ALL), and is associated with cell cycle regulation.
- Post-translational modifications include alternative initiation, isopeptide bond formation, phosphorylation, and ubiquitylation.
- Keywords associated with RGS2 include signal transduction inhibitor, translation regulation, GTPase activation, and phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen covers the full-length human RGS2 (aa 1-211); ensure that the target protein is expressed in your sample system, as tissue-specific expression may vary.
- For Western blotting, a band at ~24 kDa is expected; use appropriate controls such as known RGS2-expressing cell lines.
- In immunohistochemistry (paraffin), antigen retrieval may be necessary; optimize staining conditions for your specific tissue type.
- As a polyclonal antibody, performance may vary between batches; consider conducting validation experiments for each new lot.
- This antibody has been tested in ELISA; it may be suitable for other immunoassays, but independent validation is recommended.
