Biological Background: RGS13 Function and Localization
- RGS13 (Regulator of G protein signaling 13) is a member of the RGS protein family that negatively regulates heterotrimeric G protein signaling by accelerating the GTPase activity of G protein alpha subunits.
- It functions as a signal transduction inhibitor, driving G alpha subunits into their inactive GDP-bound state and thereby terminating downstream signaling cascades.
- RGS13 selectively binds to both G(i)-alpha and G(q)-alpha subunits, as demonstrated by similarity-based evidence.
- The protein is part of the human reference proteome and has been identified in proteomics-based studies.
- The canonical isoform (UniProt: O14921) consists of 159 amino acids with a predicted molecular mass of approximately 19.1 kDa.
Experimental Guidance and Technical Tips
- Since the immunogen is a recombinant fusion protein covering almost the entire human RGS13 sequence (amino acids 1–159), the resulting polyclonal antibody may recognize multiple epitopes on the native protein. Consider titrating the antibody to determine optimal working dilutions for your specific assays.
- For Western blot (WB), validate band specificity by using appropriate positive and negative controls, as non-specific binding can occur with polyclonal antibodies.
- The antibody is reported to cross-react with mouse and rat RGS13, making it suitable for studies in these species. However, verify performance in your own model organism.
- For ELISA, test the antibody as a capture or detection reagent under optimized buffer and blocking conditions.
