Biological Background: RGAP1 Function and Localization

  • RGAP1, also known as Rac GTPase-activating protein 1, MgcRacGAP, or CYK4, is a 71 kDa component of the centralspindlin complex, a microtubule-dependent and Rho-mediated signaling module that orchestrates contractile ring formation during cytokinesis. It sequentially binds ECT2 and RAB11FIP3 to regulate cleavage furrow ingression and abscission. Related references: PMID:18511905
  • Beyond its GAP activity, RGAP1 controls growth and differentiation of hematopoietic cells through mechanisms independent of Rac GTPase regulation. Related references: PMID:10979956
  • RGAP1 plays a critical role in erythropoiesis, and mutations in the RACGAP1 gene are associated with congenital dyserythropoietic anemia type IIIB (CDAN3B). Related references: PMID:34818416
  • RGAP1 dynamically localizes to the nucleus, cytoplasm, spindle, cleavage furrow, midbody, midbody ring, and cell membrane, reflecting its multifaceted roles during cell division.
  • Highly expressed in testis, thymus, and placenta, with lower levels in spleen and peripheral blood lymphocytes. In testis, expression is restricted to germ cells, peaking in spermatocytes. Expression is cell cycle-dependent, peaking in G2/M phase.

Experimental Guidance and Technical Tips

  • The antibody was raised against a synthetic peptide spanning amino acids 533–632; this region may be accessible in native and denatured conformations, supporting applications such as WB and IHC-P. Validate in the relevant sample system.
  • For Western blot, consider lysates from cell lines with moderate to high RGAP1 expression (e.g., HeLa, Jurkat) or testis tissue. RGAP1 expression peaks in G2/M phase; synchronization may enhance signal.
  • For IHC-P, verify staining in formalin-fixed, paraffin-embedded sections of testis, thymus, or placenta, where RGAP1 is abundantly expressed. Antigen retrieval conditions may require optimization.
  • The antibody cross-reacts with mouse and rat RGAP1; validate species-specific reactivity using appropriate tissue or cell controls.
