Biological Background: RFX3 Function and Localization
- RFX3 is a transcription factor belonging to the regulatory factor X family, with alternative splicing producing multiple isoforms.
- Essential for ciliogenesis: required for differentiation of nodal monocilia and left-right asymmetry specification, and for biogenesis of motile cilia by governing growth and beating efficiency.
- Regulates ciliary gene expression, including DYNC2LI1, FOXJ1, and BBS4 for assembly, and DNAH11, DNAH9, and DNAH5 for motility.
- Participates in endocrine pancreas development together with RFX6, driving differentiation of most islet cell types except pancreatic polypeptide-producing cells.
- Forms heterodimers with other RFX proteins and binds X-box promoter elements to regulate transcription. Related references: PMID:20148032
- Represses transcription of MAP1A specifically in non-neuronal cells, indicating cell-type-specific regulation. Related references: PMID:12411430
- Subcellular localization is strictly nuclear, consistent with its role as a transcription factor.
- Required for differentiation of ciliated ependymal cells.
Experimental Guidance and Technical Tips
- The immunogen was a recombinant fusion protein covering amino acids 280–430 of human RFX3; this region may be shared across isoforms, but validation in your specific experimental system is advised.
- For Western blotting (WB), the expected band is around 84 kDa; use appropriate positive controls (e.g., RFX3-expressing cell lines) and confirm with siRNA knockdown or overexpression.
- In ELISA applications, titrate the antibody with recombinant RFX3 protein to establish sensitivity and linear range.
- As a polyclonal antibody, batch-to-batch variability may occur; consistent lot performance should be confirmed for critical assays.
