- E3 ubiquitin-protein ligase COP1 (also known as RFWD2, RNF200, hCOP1) mediates ubiquitination and subsequent proteasomal degradation of key regulatory proteins, including p53 (TP53), JUN, SFN (14-3-3 sigma), and MTA1.
- By targeting p53 for degradation independently of MDM2 and RCHY1, COP1 abolishes p53-dependent transcription and apoptosis, promoting cell survival.
- The protein contains a RING finger domain (zinc-binding) and WD repeats, and can function as the essential RING subunit in certain E3 ligase complexes; however, in the DCX DET1-COP1 complex, the catalytic RING activity is provided by RBX1.
- Ubiquitination of SFN leads to AKT activation and enhanced cell survival, while degradation of MTA1 further underscores its role in transcriptional regulation.
- In response to TRIB1 binding, COP1 can also ubiquitinate CEBPA via a non-canonical binding motif.
- COP1 localizes predominantly to nuclear speckles and the cytoplasm, consistent with its roles in transcription factor regulation.
- It is ubiquitously expressed at low levels, with higher expression in testis, placenta, skeletal muscle, and heart.
