Biological Background: RFC1 Function and Localization
- RFC1 encodes the large subunit (140 kDa) of the heteropentameric replication factor C (RFC) complex, also known as activator 1.
- The RFC complex is required for elongation of primed DNA templates by DNA polymerases delta and epsilon, and loads PCNA onto DNA in an ATP-dependent manner (PubMed:9488738).
Related references:
PMID:9488738 - This subunit specifically binds the primer-template junction and can interact with both single- and double-stranded DNA, as well as GA-rich sequences including the PO-B transcription element, suggesting dual roles in DNA replication and transcription regulation.
- RFC1 is localized to the nucleus and is widely expressed across human tissues except placenta.
- The protein is subject to post-translational modifications, including phosphorylation and ubiquitination (isopeptide bond), as indicated by large-scale proteomic studies.
- Mutations in RFC1 are associated with cerebellar ataxia, neuropathy, vestibular areflexia syndrome (CANVAS), expanding its clinical relevance beyond core DNA metabolism.
- Alternative splicing generates multiple transcript variants, contributing to functional diversity.
Experimental Guidance and Technical Tips
- For Western blot, expect a band around 128 kDa. Use 6-8% SDS-PAGE for optimal separation of high-molecular-weight proteins.
- For immunofluorescence/ICC, include a nuclear counterstain (e.g., DAPI) and confirm nuclear localization. Permeabilization conditions may need optimization.
- ELISA: The antibody may be used to detect recombinant or endogenous RFC1; titrate appropriately.
- Sequence alignment of the immunogen (aa 400-700) across human, mouse, and rat indicates high conservation, supporting reported cross-reactivity; however, validate in your specific experimental setup.
