Biological Background: REXO2 Function and Localization
- REXO2 is a 3'-to-5' exoribonuclease that preferentially degrades DNA and RNA oligonucleotides composed of only two nucleotides, but binds and degrades longer oligonucleotides with lower affinity. Related references: PMID:23741365, PMID:30926754, PMID:31588022, PMID:32365187
- In mitochondria, REXO2 scavenges nanoRNAs produced by the degradosome and clears short RNAs generated by RNA processing, and is essential for correct initiation of mitochondrial transcription by degrading mitochondrial RNA dinucleotides to prevent RNA-primed transcription at non-canonical sites. Related references: PMID:30926754, PMID:31588022, PMID:32365187
- Alternative names include SFN, REX2, CGI-114, and small fragment nuclease.
- Subcellular localization is primarily mitochondrial (intermembrane space and matrix), with additional presence in the cytoplasm and nucleus.
- Post-translational modifications include acetylation and phosphorylation, as identified by proteomics.
- Highly expressed in the heart, with lower expression in lymph nodes, brain, lung, liver, spleen, and thymus.
- Essential for embryonic development, as shown by similarity to other organisms.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 26-237 of human REXO2, representing the mature protein after cleavage of the predicted mitochondrial transit peptide; this design likely produces antibodies that recognize both precursor and mature forms.
- For Western blot, a specific band near 27 kDa is expected. Due to potential post-translational modifications or processing, multiple bands may appear; validate in your sample system using appropriate controls.
- For immunofluorescence (IF/ICC), consider REXO2's primary mitochondrial localization; co-stain with a mitochondrial marker to confirm specificity, and note possible cytoplasmic and nuclear signals.
- For ELISA, the antibody can be used for capture or detection. Optimize coating concentrations and detection conditions empirically.
- As a polyclonal antibody, lot-to-lot variation may occur; perform lot-specific validation for critical or quantitative experiments.
