Biological Background: Ret Function and Localization

  • Ret is a receptor tyrosine kinase encoded by the RET proto-oncogene (also known as CDHF12, CDHR16, PTC) and belongs to the cadherin superfamily.
  • Receptor activation requires binding of GDNF family ligands (GDNF, NRTN, ARTN, PSPN, GDF15) to specific GDNFR-α coreceptors, leading to autophosphorylation and downstream MAPK and AKT signaling. Related references: PMID:21994944, PMID:23333276, PMID:24560924
  • Functions as a dependence receptor in pituitary somatotrophs, promoting survival in the presence of GDNF but triggering apoptosis upon ligand withdrawal. Related references: PMID:20616503, PMID:21994944
  • Involved in organogenesis: required for development of the enteric nervous system, kidneys, and gut-associated lymphoid tissues (by similarity), and triggers differentiation of rapidly adapting mechanoreceptors. Related references: PMID:20064382
  • Mediates GDF15-induced aversive responses in the brainstem, including nausea and appetite loss, via the GFRAL coreceptor. Related references: PMID:28846097, PMID:28846099, PMID:28953886
  • Regulates cell adhesion and migration through caspase-mediated cleavage and integrin-dependent mechanisms. Related references: PMID:20702524, PMID:21357690
  • Phosphorylates PTK2/FAK1 upon activation. Related references: PMID:21454698
  • Alternative splicing generates isoforms with differential signaling potency; isoform 1 induces stronger MAPK activation than isoform 2. Mutations in RET are associated with multiple endocrine neoplasia type 2 (MEN2A/MEN2B) and Hirschsprung disease.

Experimental Guidance and Technical Tips

  • The immunogen targets a C-terminal peptide (aa 1000–1100) within the kinase domain; thus the antibody may recognize full-length Ret (~124 kDa) and possibly C-terminal fragments or cleaved isoforms.
  • For Western blotting, consider using reducing conditions and validate the expected molecular weight; be aware that glycosylation and proteolytic processing may produce additional bands.
  • In immunofluorescence/ICC, anticipate membrane and endosomal staining; optimize fixation and permeabilization protocols for your cell type.
  • Include appropriate controls such as known Ret-expressing cell lines (e.g., neuroendocrine or thyroid cancer cells) and imatinib-treated samples to confirm specificity.
  • For ELISA applications, titrate the antibody and use a matched pair strategy with a detection antibody for optimal sensitivity.
