Biological Background: REST/NRSF Function and Localization

  • REST/NRSF, also known as RE1-silencing transcription factor or neural-restrictive silencer factor, is a transcriptional repressor that binds to neuron-restrictive silencer elements (NRSE) to silence neuronal genes in non-neuronal cells. It recruits corepressors SIN3A and RCOR1, which in turn attract histone deacetylases to modify chromatin and enforce repression. Related references: PMID:11741002, PMID:11779185, PMID:12399542, PMID:10449787, PMID:10734093
  • REST recruits the BHC complex for histone deacetylation and demethylation, and can form a repressive complex with CDYL and histone methyltransferase EHMT2, which is implicated in transformation suppression. Related references: PMID:19061646
  • Isoform 3 (a shorter splice variant) exhibits weaker repressor activity and may form heterodimers with full-length REST, inhibiting its DNA binding and corepressor recruitment, thereby derepressing target genes. This isoform switching is crucial for hearing and neuronal gene activation. Related references: PMID:11779185, PMID:11741002
  • REST maintains the quiescent state of adult neural stem cells by repressing neuronal genes; its dissociation from NRSE sites permits differentiation into mature neurons. Related references: PMID:21258371
  • During postnatal development, REST represses GRIN2B expression, facilitating a switch in synaptic NMDA receptor subunit composition from GRIN2B to GRIN2A (By similarity).
  • Under hypoxic conditions, REST acts as a key repressor by directly binding to RE1/NRSE sites in promoters of oxygen‑sensitive genes. Related references: PMID:27531581
  • REST may confer stress resistance in the aging brain by regulating genes involved in cell death and stress responses. Related references: PMID:24670762
  • In breast cancer, REST negatively regulates the expression of SRRM3, and loss of REST correlates with increased invasiveness. Related references: PMID:26053433
  • Ubiquitously expressed, with higher levels in lymphoid tissues; in neurons of the prefrontal cortex and hippocampus, nuclear REST decreases in Alzheimer disease, frontotemporal dementia, and Parkinson disease, often mislocalizing to Lewy bodies. Related references: PMID:24670762, PMID:30684677, PMID:21258371, PMID:26053433, PMID:8568247
  • REST localizes to both the nucleus and cytoplasm, contains nine C2H2‑type zinc finger motifs, and is regulated by phosphorylation and ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen (residues 760–1060) lies within the C‑terminal region of full‑length REST (isoform 1). This antibody is expected to recognize isoform 1; the shorter isoform 3 may not be detected. Validate reactivity in the relevant sample system.
  • For IF/ICC, optimize fixation and permeabilization conditions. Nuclear staining should predominate, but cytoplasmic signal may be observed under certain conditions.
  • Choose appropriate cell models: non‑neuronal lines (e.g., HEK293, HeLa) express REST, while mature neurons typically show low or absent expression. Use siRNA knockdown or peptide blocking to confirm specificity.
  • For ELISA, the immunogen sequence or full‑length recombinant REST can serve as a standard or blocking peptide. Consider both direct and indirect assay formats.
  • As a polyclonal antibody, verify lot‑to‑lot consistency when transitioning between batches for critical experiments.
