Biological Background: RELB Function and Localization
- RELB (transcription factor RelB, also known as I-Rel) is a member of the NF-κB family of transcription factors, encoded by the RELB gene in humans. It functions as a DNA-binding activator or repressor in homo- or heterodimeric complexes.
- NF-κB dimers containing RELB (e.g., RelB-p50, RelB-p52) bind to κB sites in target gene promoters, playing critical roles in inflammation, immunity, cell growth, differentiation, and apoptosis.

- It contributes to pancreatic ductal adenocarcinoma resistance by participating in the NUPR1/RELB/IER3 survival pathway under cell stress conditions.
- RELB regulates the circadian clock by repressing the transcriptional activator activity of the CLOCK-BMAL1 heterodimer, independently of CRY proteins, with enhanced repression in the presence of NFKB2/p52.
