Biological Background: REG4 Function and Localization
- Also known as GISP, RELP, REG-IV, Reg IV, this protein is a member of the regenerating islet-derived protein family.
- Functions as a calcium-independent lectin with mannose-binding specificity, capable of maintaining carbohydrate recognition in acidic environments.
- May play a role in inflammatory and metaplastic responses of the gastrointestinal epithelium.
- Highly expressed in the gastrointestinal tract, including duodenum, jejunum, ileum, ileocecum, appendix, descending colon, pancreas, and small intestine; weakly expressed in normal colon and stomach.
- Strongly upregulated in most colorectal tumors compared to normal colon, with preferential expression in mucinous and neuro-endocrine tumors; localized to mucus-secreting cells and enterocyte-like cells.
- Contains a signal peptide and is secreted; post-translational modifications include glycosylation and disulfide bond formation.

- The immunogen corresponds to amino acids 23–158 of human REG4, covering the majority of the mature protein; the antibody may detect endogenous REG4 in western blot (WB) at ~18 kDa.
- For WB, consider optimizing protein loading and antibody dilution, and validate in colorectal cancer cell lines or tissue lysates where REG4 is highly expressed.
- The antibody cross-reacts with mouse and rat REG4, making it suitable for multi-species studies; however, confirm cross-reactivity in your specific experimental system.
