Biological Background: REG3A Function and Localization
- REG3A (Regenerating islet-derived protein 3-alpha), also known as hepatointestinal pancreatic protein (HIP/PAP) or pancreatitis-associated protein 1, is a bactericidal C-type lectin.
- It acts exclusively against Gram-positive bacteria by binding to surface-exposed carbohydrate moieties of peptidoglycan and killing bacteria by forming a hexameric membrane-permeabilizing oligomeric pore (PubMed:16931762, PubMed:24256734).
Related references: PMID:16931762, PMID:24256734 - REG3A functions as a hormone, activating the EXTL3 receptor to trigger cell-specific signaling pathways (PubMed:19158046, PubMed:22727489, PubMed:27830702).
Related references: PMID:19158046, PMID:22727489, PMID:27830702 - It regulates keratinocyte proliferation and differentiation after skin injury via the EXTL3-PI3K-AKT pathway and inhibits skin inflammation by suppressing IL-6 and TNF (PubMed:22727489, PubMed:27830702).
Related references: PMID:22727489, PMID:27830702 - In the pancreas, REG3A permeabilizes beta-cell membranes and stimulates their proliferation (PubMed:19158046).
Related references: PMID:19158046 - The protein is secreted and contains disulfide bonds typical of the C-type lectin family.
- Tissue expression is constitutive in the intestine, low in healthy pancreas, but highly upregulated in psoriasis keratinocytes and during acute pancreatitis (PubMed:22727489, PubMed:1469087).
Related references: PMID:22727489, PMID:1469087 - REG3A also exhibits bacteriostatic activity.
Experimental Guidance and Technical Tips
- For Western blot (WB), the predicted molecular weight is 19 kDa. Include appropriate positive and negative controls, and validate band specificity in your target tissue or cell line.
- In immunohistochemistry (IHC-P), antigen retrieval may be required. Test on known positive tissues such as inflamed pancreas or psoriatic skin.
- For ELISA, consider pairing with a detection antibody specific to REG3A; optimize coating concentrations for your assay format.
- Due to the polyclonal nature, some lot-to-lot variation may occur; we recommend titrating each new lot and including internal reference standards.
- Cross-reactivity with mouse and rat is indicated; however, we suggest verifying species-specific reactivity in your experimental system.
