Biological Background: RECK Function and Localization
- RECK (reversion-inducing cysteine-rich protein with Kazal motifs), also known as suppressor of tumorigenicity 15 protein (ST15), is a GPI-anchored cell membrane glycoprotein with serine protease inhibitor domains.
- Functions as a Wnt7-specific coactivator of canonical Wnt signaling, interacting with the disordered linker of Wnt7 to confer ligand selectivity; forms a complex with ADGRA2, Fzd, LRP5/6. Related references: PMID:30026314
- Together with ADGRA2, enables brain endothelial cells to respond to Wnt7 signals, essential for CNS angiogenesis and blood-brain barrier regulation. Related references: PMID:28289266, PMID:30026314
- Acts as a broad-spectrum metalloproteinase inhibitor, suppressing MMP9 secretion and directly inhibiting MMP9, MMP2, MMP14 (MT1-MMP), and ADAM10 activities. Related references: PMID:18194466, PMID:9789069, PMID:33731436
- Expressed in various normal tissues and untransformed cells; expression is lost in many tumor-derived and oncogenically transformed cell lines, consistent with its role as a tumor suppressor. Related references: PMID:9789069
- Contains multiple Kazal-type serine protease inhibitor repeats and a GPI-anchor, and is classified under keywords: 3D-structure, cell membrane, disulfide bond, glycoprotein, GPI-anchor, lipoprotein, membrane, protease inhibitor, proteomics identification, reference proteome, repeat, serine protease inhibitor, signal, tumor suppressor, Wnt signaling pathway.
- The full-length protein has a predicted molecular weight of approximately 106 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 110–210 of human RECK, a region that may include one or more Kazal motifs. For Western blot, consider using membrane protein extraction protocols and possibly reducing versus non-reducing conditions to optimize detection, given the protein’s GPI-anchored nature and disulfide bonds.
- In ELISA, the recombinant immunogen can be utilized as a coating antigen. Validate in the relevant sample system, and perform titration to determine optimal antibody and antigen concentrations.
- When studying species cross-reactivity, verify performance in mouse samples, as the antibody has been reported to cross-react with mouse RECK.
- For applications beyond WB and ELISA, pilot experiments are recommended; the antibody may work in other immunoassays if the epitope is accessible.
