Biological Background: Roquin-1 Function and Localization
- RC3H1 encodes Roquin-1, a member of the RING finger and CCCH-type zinc finger protein family, also known as RNF198.
- It functions as a post-transcriptional repressor by binding to constitutive decay elements (CDE) in the 3'-UTR of target mRNAs (e.g., ICOS, TNF, TNFRSF4), promoting their deadenylation and degradation. Related references: PMID:25026078, PMID:31636267
- In follicular helper T cells, Roquin-1 represses ICOS and TNFRSF4 expression, preventing spontaneous Tfh cell differentiation and autoimmunity.
