Biological Background: RBPJK Function and Localization

  • Transcriptional regulator that plays a central role in Notch signaling, a pathway involved in cell-fate determinations. Acts as a repressor in the absence of Notch and as an activator when bound to the Notch intracellular domain (NICD). Related references: PMID:41086914
  • Recruits chromatin remodeling complexes containing histone deacetylase or histone acetylase proteins to repress or activate transcription, respectively.
  • Specifically binds to the immunoglobulin kappa-type J segment recombination signal sequence.
  • Recognizes and binds specifically to methylated DNA. Related references: PMID:21991380
  • Activates the transcription of COX4I2 under hypoxia conditions (4% oxygen) by binding to its oxygen responsive element. Related references: PMID:23303788
  • Negatively regulates the phagocyte oxidative burst in response to bacterial infection by repressing transcription of NADPH oxidase subunits (By similarity).
  • Predominantly localizes to the nucleus but can also be found in the cytoplasm.
  • Exists in multiple isoforms due to alternative splicing and is associated with disease variants; also involved in host-virus interactions.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1-260) of human RBPJK; consider that the antibody may recognize multiple splicing isoforms.
  • For Western blot, expect a band around 56 kDa; validate in the relevant sample system and optimize loading controls.
  • For IHC-P, standard antigen retrieval and appropriate blocking steps are recommended; pilot staining with known positive and negative tissues is advised.
  • In ELISA, the antibody can be used for capture or detection, but optimal coating concentration and detection conditions should be determined empirically.
  • Cross-reactivity with mouse and rat has been reported, but confirm expression and sequence homology in your specific experimental context.
