Biological Background: RBP4 Function and Localization
- Retinol-binding protein 4 (RBP4), also known as plasma retinol-binding protein (PRBP or RBP), is a secreted carrier of the lipocalin family that specifically transports retinol (vitamin A) in blood plasma. Related references: PMID:5541771
- Once loaded with retinol in the liver, RBP4 circulates and delivers the vitamin to peripheral tissues, playing an essential role in vision, cellular differentiation, and vitamin A homeostasis.
- RBP4 transfers its bound all-trans retinol to the STRA6 receptor on target cells, which then facilitates retinol transport across the cell membrane. Related references: PMID:22665496
- The protein is secreted and detected in blood plasma and urine, consistent with its transport function and renal clearance.
- RBP4 contains a single disulfide bond and undergoes post-translational methylation; mutations in RBP4 are associated with microphthalmia, underscoring its critical role in ocular development.
- The mature protein has a calculated molecular weight of approximately 23 kDa, and typically migrates around 21 kDa under reducing SDS-PAGE conditions.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 19–201 of human RBP4, corresponding to the mature secreted form, so the antibody is expected to recognize both full-length and processed protein.
- For Western blot, consider using human plasma, serum, or liver lysates as positive controls; a band near 21 kDa may be observed. Optimize primary antibody dilution and detection conditions empirically.
- For ELISA, direct or sandwich formats may be tested; titrate the antibody against recombinant RBP4 or reference serum samples to determine optimal working concentrations.
- The antibody shows cross-reactivity with mouse RBP4, making it suitable for studies in mouse models, but validation in the relevant sample type is recommended.
- To maintain stability, aliquot and store at -20°C or -80°C, avoiding repeated freeze-thaw cycles.
