Biological Background: RBM39 Function and Localization
- RNA-binding protein 39 (RBM39), also known as CAPERα or HCC1, is a pre-mRNA splicing factor that promotes exon inclusion via regulation of exon cassette splicing. Related references: PMID:15694343, PMID:24795046, PMID:28302793, PMID:31271494
- Besides its splicing role, RBM39 functions as a transcriptional coactivator for steroid nuclear receptors ESR1/ER-alpha and ESR2/ER-beta, and JUN/AP-1, independently of its splicing activity.
- The protein localizes to nuclear speckles, consistent with its involvement in mRNA processing.
- RBM39 is widely expressed, with highest levels in pancreas, skeletal muscle, lung, and brain, and intermediate levels in kidney, liver, and heart.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitin-like (Ubl) conjugation, suggesting complex regulation.
- The protein contains RNA-binding motifs and is involved in alternative splicing, contributing to proteome diversity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 400-508 of human RBM39, so the antibody is expected to recognize epitopes within this region; consider validating reactivity in your specific experimental system.
- For Western blot, a band around 59 kDa is expected; use appropriate controls and optimize antibody dilution.
- For IHC-P and IF/ICC, nuclear speckle localization should be observed; include proper fixation and antigen retrieval steps.
- Cross-reactivity with mouse and rat is reported; validate in those species if using non-human samples.
- For ELISA, the antibody can be used in direct or indirect formats; the immunogen peptide may serve as a positive control.
- Always validate antibody performance in your specific sample type and assay conditions before routine use.
