- Splicing factor 45 (RBM17), also known as RNA-binding motif protein 17, is a ~45 kDa protein that functions in pre-mRNA splicing as part of the spliceosome.
- Binds to the single-stranded 3'AG at the exon/intron border and promotes its utilization in the second catalytic step of splicing.
- Plays a role in alternative splicing regulation and can promote the use of cryptic splice sites. For instance, it facilitates a cryptic splice site generated by the beta-110 mutation in the HBB gene, which leads to sickle cell anemia.
- Localizes predominantly to the nucleus, consistent with its role in mRNA processing.
- Subject to post-translational modifications, including acetylation, phosphorylation, and ubiquitination (isopeptide bond formation).
- Identified as a proteomics target and reference proteome member with RNA-binding activity.

- The immunogen spans amino acids 1-240 of human RBM17 (N-terminal region), which should recognize both full-length and truncated forms of the protein.
- For Western blot, expect a band around 45 kDa in human, mouse, and rat cell or tissue lysates. Consider using appropriate loading controls.
