Biological Background: RBM14 Function and Localization
- RBM14 (RNA-binding protein 14), also known as COAA, PSP2, and SYTIP1, is a nuclear RNA-binding protein with two major isoforms arising from alternative splicing.
- Isoform 1 functions as a transcriptional coactivator via NCOA6 and CITED1, while isoform 2 acts as a repressor, modulating transcription. Related references: PMID:11443112

Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against a recombinant protein spanning amino acids 110–220 of human RBM14. It may recognize both isoforms in human, mouse, and rat samples, but cross-reactivity with other species should be independently confirmed.
- For Western blot, a band near 69 kDa is expected in lysates from cell lines or tissues expressing RBM14. Optimize primary antibody dilution (e.g., 1:500–1:2000) and use appropriate blocking conditions; validate with positive and negative controls.
- In immunofluorescence/ICC, expect nuclear and nucleolar staining, potentially with weak cytoplasmic signal. Include nuclear counterstains and test permeabilization conditions.
- For ELISA applications, the antibody can be used as a detection or capture reagent; titrate to determine optimal working concentration. Consider pairing with a different clone for sandwich ELISA.
