Biological Background: RBBP5 Function and Localization
- As a scaffold protein, RBBP5 forms the WRAD subcomplex with WDR5, ASH2L, and DPY30, essential for the full catalytic activity of KMT2 enzymes (MLL1-4, SETD1A/B) in methylating histone H3 at lysine 4. Related references: PMID:21220120, PMID:22266653
- In embryonic stem cells, RBBP5 is critical for differentiation potential along the neural lineage by regulating gene induction and H3K4 methylation at developmental loci, including those activated by retinoic acid (By similarity).
- As part of the MLL1/MLL complex, RBBP5 is required for mono-, di-, and trimethylation of histone H3 at lysine 4 (H3K4me1/2/3), a hall mark of active transcription. Related references: PMID:19131338, PMID:19556245
- RBBP5 is predominantly localized in the nucleus, consistent with its role in chromatin modification and transcriptional regulation.
- It is ubiquitously expressed across human tissues, reflecting its fundamental role in global H3K4 methylation.

- Based on the immunogen design (aa 319-538), this antibody is expected to recognize the C-terminal region of RBBP5. For Western blot, a band near 59 kDa is predicted; optimize loading and transfer conditions.
- In immunofluorescence and immunohistochemistry, nuclear staining should be observed. Ensure proper permeabilization and antigen retrieval protocols.
- For ELISA, validate the assay with an appropriate coating antigen; cross-reactivity with other proteins in complex samples should be independently tested.
