Biological Background: RBBP4 Function and Localization
- RBBP4, also known as NURF55, RBAP48, p48, is a core histone-binding subunit belonging to the WD repeat protein family.
- It is a component of the chromatin assembly factor 1 (CAF-1) complex, which facilitates nucleosome assembly on newly replicated and repaired DNA. Related references: PMID:8858152
- RBBP4 is part of multiple histone deacetylase complexes, including the core HDAC complex and the NuRD complex, promoting transcriptional repression via histone deacetylation and nucleosome remodeling. Related references: PMID:9150135 PMID:16428440 PMID:28977666 PMID:39460621
- It is also a subunit of the NURF (nucleosome remodeling factor) complex, connecting chromatin remodeling to transcriptional regulation. Related references: PMID:14609955 PMID:15310751
- As a component of the PRC2 complex, RBBP4 contributes to the repression of homeotic genes during development. Related references: PMID:29499137 PMID:31959557
- RBBP4 localizes to the nucleus, and is found at chromosome telomeres, consistent with its roles in chromatin modification.
- It is expressed in neuroblastoma cells, and undergoes post-translational modifications including acetylation, phosphorylation, and ubiquitination.
- Keywords: Cell cycle, DNA damage, DNA repair, DNA replication, Transcription regulation, WD repeat.
Experimental Guidance and Technical Tips
- The recombinant full-length immunogen suggests the antibody may detect native RBBP4 protein; consider testing in relevant sample systems.
- For Western blot analysis, use reducing and denaturing conditions, and expect a band at approximately 48 kDa consistent with the protein weight.
- This antibody is reported for ChIP; appropriate crosslinking and chromatin preparation protocols should be optimized for your specific experimental system.
- ELISA can be used for quantitative detection; validate linearity and sensitivity with purified protein standards.
