Biological Background: RASSF5 Function and Localization
- RASSF5, also known as NORE1, NORE1A, NORE1B, RASSF3, Maxp1, or RAPL, is a potential tumor suppressor that is frequently down-regulated in lung tumor cell lines and primary lung tumors.
- Isoform 2 of RASSF5 plays a key role in lymphocyte adhesion by linking RAP1A activation upon T-cell receptor or chemokine stimulation to integrin activation, stimulating lymphocyte polarization and enhancing adhesion to ICAM1.
- In endothelial cells, the association of RASSF5 isoform 2 with activated RAP1A is required for directional movement during wound healing.
- RASSF5, together with RAP1A, may participate in the regulation of microtubule growth.
- The RASSF5-STK4/MST1 complex may mediate HRAS and KRAS-induced apoptosis.
- RASSF5 localizes to the cytoplasm and associates with the cytoskeleton.
- The protein contains a zinc-finger domain, undergoes acetylation and phosphorylation, and is annotated with keywords including alternative splicing, apoptosis, and tumor suppressor.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 1-265 amino acids of human RASSF5 (NP_872606.1). When using this antibody, consider that it may recognize multiple isoforms of RASSF5 (the region spans part of the protein), including the widely expressed isoform 1 and isoform 2. Validate specificity in your sample system.
- For Western blotting, the expected molecular weight of RASSF5 is approximately 47 kDa. Use appropriate loading controls to confirm band identity, as post-translational modifications may cause slight shifts.
- The antibody has been shown to cross-react with mouse samples; however, always verify cross-reactivity in your specific tissue or cell type, as expression levels may vary.
- For ELISA, optimize coating antigen concentration and antibody dilution to achieve optimal signal-to-noise ratio, considering the polyclonal nature of the antibody.
