Biological Background: RASSF1 Function and Localization
- RASSF1 is a potential tumor suppressor required for death receptor-dependent apoptosis. It mediates activation of STK3/MST2 and STK4/MST1 during Fas-induced apoptosis by preventing their dephosphorylation.
- Upon association with MOAP1, RASSF1 promotes BAX conformational change and translocation to mitochondrial membranes in response to TNF and TNFSF10 stimulation.
- Isoform A interacts with CDC20, an activator of the anaphase-promoting complex (APC), inhibiting APC activity and mitotic progression. It also inhibits proliferation by regulating cyclin D1 accumulation at the G1/S-phase transition.
- Isoform A disrupts the MDM2-DAXX-USP7 interaction, promoting MDM2 self-ubiquitination and contributing to efficient TP53 activation in cell-cycle checkpoint control following DNA damage.
- The protein localizes to the cytoplasm, cytoskeleton (including microtubules, centrosome, spindle, and spindle pole), and nucleus.
- Isoforms A and C are ubiquitously expressed in all tissues tested, but isoform A is frequently absent in cancer cell lines. Isoform B is mainly expressed in hematopoietic cells.
- Key post-translational modifications include acetylation, methylation, and phosphorylation. The protein contains a zinc-finger domain and binds zinc.
- Alternative splicing and alternative promoter usage produce multiple isoforms, and the gene is subject to epigenetic silencing in many cancers.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 101–340 of human RASSF1 (isoform A), encompassing critical functional domains. Consider verifying isoform specificity using positive controls expressing known RASSF1 variants.
- For Western blot analysis, whole cell lysates from human or mouse tissues/cell lines expressing RASSF1 should yield a band near 39 kDa. Note that post-translational modifications may cause slight mobility shifts.
- In ELISA, using the recombinant protein or peptide derived from the immunogen region as coating antigen can help confirm antibody recognition. Validate assay conditions with appropriate positive and negative controls.
- Given that RASSF1 is frequently silenced by promoter methylation in cancer, treatment with demethylating agents (e.g., 5-aza-2'-deoxycytidine) before lysate preparation may be necessary for detection in certain cell lines.
- This antibody has been shown to react with human and mouse samples; cross-reactivity with other species should be determined empirically.
