Biological Background: RASGRP1 Function and Localization

  • RASGRP1 (RAS guanyl-releasing protein 1), also known as CalDAG-GEFII (Calcium and DAG-regulated guanine nucleotide exchange factor II), is a Ras-specific guanine nucleotide exchange factor that activates Ras by catalyzing GDP/GTP exchange.
  • It functions as a key downstream effector of T-cell and B-cell antigen receptors, coupling receptor activation to the Ras-ERK/MAP kinase cascade, thereby regulating lymphocyte development, homeostasis, and differentiation.
  • RASGRP1 is essential for NK cell cytotoxicity and ITAM-dependent cytokine production, activating both ERK and JNK pathways.

  • The immunogen spans amino acids 678–795 in the C-terminal region of mouse RASGRP1; this region is conserved between mouse and human, supporting cross-reactivity.
  • For Western blotting, a predominant band near 90 kDa may be observed; consider validating lysates from spleen, thymus, or immune cell lines.
  • For immunoprecipitation, use standard protocol conditions; the polyclonal nature may enrich target effectively; verify with protein A/G agarose.
  • In ELISA, use purified recombinant protein or cell lysates as coating antigen to assess binding.
  • As a rabbit polyclonal antibody, it recognizes multiple epitopes; specificity should be confirmed in knockout or knockdown systems.
