  • RASA1, also known as Ras p21 protein activator or p120GAP, is a GTPase-activating protein (GAP) that stimulates the intrinsic GTPase activity of Ras proteins such as NRAS, promoting their inactivation.
  • By facilitating the transition of Ras from the active GTP-bound to the inactive GDP-bound state, RASA1 acts as a negative regulator of Ras-mediated signal transduction pathways.
  • The protein contains SH2 and SH3 domains, as well as repeat regions, which mediate protein-protein interactions and contribute to its regulatory function.
  • RASA1 is a cytoplasmic protein and is recruited to the plasma membrane where it interacts with activated Ras to terminate downstream signaling.
  • In placental villi, RASA1 expression is detected exclusively in trophoblast cells (cytotrophoblast and syncytiotrophoblast) and is absent from stromal, endothelial, and Hofbauer cells.
