Biological Background: RARβ Function and Localization
- RARβ (retinoic acid receptor beta, also known as HBV-activated protein, NR1B2, RAR-epsilon) is a member of the nuclear receptor superfamily that mediates the effects of retinoic acid, a key morphogen involved in development and cellular differentiation.
- It forms heterodimers with retinoid X receptors (RXR) and binds to specific DNA sequences called retinoic acid response elements (RARE), composed of tandem AGGTCA motifs, to regulate transcription of target genes.
- RARβ acts primarily as a transcriptional activator due to weak interaction with corepressors. Related references: PMID:12554770
- Depending on the response element architecture, the RXR/RARβ heterodimer can switch between activation (on DR5 elements) and repression (on DR1 elements), providing fine-tuned control of gene expression. Related references: PMID:29021580
- Together with RARG, RARβ is essential for skeletal development, growth plate function, and matrix homeostasis.
- The protein localizes to both the nucleus and cytoplasm, consistent with its role as a transcription factor and potential non-genomic functions.
- Expression is detected at the protein level in aortic endothelial cells, implicating roles in vascular biology.
- RARβ undergoes phosphorylation as a post-translational modification and contains zinc-finger motifs that are critical for DNA binding and transcriptional regulation.
Experimental Guidance and Technical Tips
- This antibody was raised against a peptide mapping to amino acids 350-455 of human RARβ; due to alternative splicing, verify the detected band size (~50 kDa) by western blotting in your sample system and consider appropriate controls if cross-reacting with mouse tissues.
- For immunofluorescence (IF/ICC), optimize fixation and permeabilization conditions to improve nuclear signal detection.
- In ELISA, ensure optimal coating antigen concentration and detection parameters to achieve reliable quantification.
