Biological Background: RARRES2 Function and Localization
- RARRES2 (chemerin) is a secreted adipokine encoded by the RARRES2 gene, also known as TIG2 or tazarotene-induced gene 2 protein.
- It regulates adipogenesis, metabolism, and inflammation through activation of the chemokine-like receptor 1 (CMKLR1), and also acts as a ligand for CMKLR2. It can additionally bind CCRL2 with lower affinity. Related references: PMID:27716822
- Positively regulates adipocyte differentiation and modulates the expression of genes involved in lipid and glucose metabolism; may play a role in angiogenesis during white adipose tissue expansion.
- Exhibits dual pro- and anti-inflammatory properties depending on proteolytic cleavage; acts as a chemotactic factor for dendritic cells, macrophages, and NK cells.
- In vascular endothelial cells, exerts anti-inflammatory effects by preventing TNF-induced VCAM1 expression and monocyte adhesion via AKT1/NOS3 signaling and nitric oxide production.
- Highly expressed in placenta, liver, white adipose tissue (enriched in adipocytes), and skin (epidermis, hair follicles, endothelial cells).
- Expression elevated in metabolic and inflammatory diseases such as psoriasis, obesity, type 2 diabetes, metabolic syndrome, and cardiovascular disease.
- Displays antimicrobial activity in skin; contains a signal peptide and disulfide bonds.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 17-163 of human RARRES2, covering nearly the full-length secreted protein. For Western blot, a band may be observed near 19 kDa.
- For immunofluorescence/immunocytochemistry, consider that RARRES2 is a secreted protein; intracellular staining may require permeabilization and stimulation of release.
- Validate antibody performance in appropriate positive control samples (e.g., human adipose tissue or liver lysates) and consider species cross-reactivity with mouse samples as indicated.
- When performing ELISA, pair this polyclonal antibody with a suitable detection antibody; optimize conditions for your specific assay format.
