- RAPGEF1 (also known as C3G, GRF2) is a guanine-nucleotide-releasing protein that binds to the SH3 domain of CRK and GRB2/ASH, transducing signals from CRK to activate RAS.
- It is involved in cell branching and adhesion through the BCAR1-CRK-RAPGEF1 signaling cascade, leading to activation of RAP1. Related references: PMID:12432078
- Plays a role in the establishment of basal endothelial barrier function, contributing to vascular integrity.
- Mediates nerve growth factor (NGF)-induced sustained activation of Rap1 and promotes neurite outgrowth, highlighting its importance in neurogenesis.
- The protein localizes primarily to early endosomes, suggesting involvement in endosomal signaling.
- RAPGEF1 is ubiquitously expressed in adult and fetal tissues, with high expression in adult skeletal muscle and placenta, and in fetal brain and heart; expression is low in adult and fetal liver.
