Biological Background: Rap1A Function and Localization
- Rap1A, or Ras-related protein 1A, is a small GTPase of the Ras superfamily, also known as C21KG, G-22K, GTP-binding protein smg p21A, and Krev-1.
- It counteracts the mitogenic function of Ras by competitively interacting with Ras GAPs and RAF. Related references: PMID:17916086
- Together with ITGB1BP1, Rap1A regulates KRIT1 localization to microtubules and membranes. Related references: PMID:17916086
- Plays a role in nerve growth factor (NGF)-induced neurite outgrowth.
- Involved in the regulation of embryonic blood vessel formation and establishment of basal endothelial barrier function.
- Facilitates the progressive accumulation of CDH1 at mature desmosome junctions via cAMP-dependent signaling and its interaction with PKP3. Related references: PMID:25208567
- Subcellular localization includes the cell membrane, cytoplasm (perinuclear region), cell junctions, and early endosomes.
- Key features: lipoprotein, prenylated, methylated, GTP-binding, and hydrolase activity; classified as a tumor suppressor.
Experimental Guidance and Technical Tips
- The immunogen targets a sequence within amino acids 39–138, covering a portion of the conserved GTPase domain. Consider performing a BLAST alignment to assess potential cross-reactivity with other Ras family members.
- For Western blot, a band around 21 kDa is expected; include appropriate molecular weight markers and validate using knockout or knockdown samples where possible.
- For immunohistochemistry (IHC-P), optimize antigen retrieval conditions and confirm specificity using tissue sections from knockout animals if available.
- The antibody has been validated for knockout in WB; however, performance in other applications should be evaluated in your specific experimental system.
