Biological Background: RanBP9 Function and Localization

  • RanBP9 (Ran-binding protein 9, also known as RanBPM, BPM-L, BPM90, RanBP7) is a scaffolding protein that may act as an adaptor to couple membrane receptors to intracellular signaling pathways.
  • It is a core component of the CTLH E3 ubiquitin-protein ligase complex, mediating ubiquitination and subsequent proteasomal degradation of target proteins such as HBP1. Related references: PMID:29911972
  • RanBP9 enhances HGF-MET signaling by recruiting Sos and activating the Ras pathway, and may be involved in integrin signaling, including ITGB2/LFA-1. Related references: PMID:12147692 PMID:14722085

  • For Western blot (WB), this polyclonal antibody, raised against a recombinant immunogen spanning amino acids 480–605, may recognize linear epitopes. A band of approximately 78 kDa is expected in human cell lysates; consider using a positive control such as HeLa or HEK293 whole cell extract.
  • Due to potential isoform variations from alternative splicing, confirm the molecular weight and specificity in your target tissue or cell line.
  • For ELISA, the antibody can be considered for direct detection of recombinant or native RanBP9; however, optimal coating concentration and detection conditions should be empirically determined.
  • As cross-reactivity is reported for human, validate performance in other species of interest before use.
