Biological Background: Ran Function and Localization
- Ran (TC4, Gsp1, ARA24) is a small GTPase belonging to the Ras superfamily, encoded by the RAN gene in humans.
- Functions as a key regulator of nucleocytoplasmic transport, controlling the import and export of proteins and RNAs by cycling between GDP- and GTP-bound states; this cycling ensures directional cargo loading and release by transport receptors. Related references: PMID:10400640, PMID:17209048, PMID:26272610, PMID:11336674, PMID:29040603
- Essential for mitosis: in its GTP-bound form, Ran triggers microtubule assembly at mitotic chromosomes and is required for normal spindle formation and chromosome segregation; interacts with BIRC5/survivin to deliver TPX2 to microtubules. Related references: PMID:10408446, PMID:29040603, PMID:12194828, PMID:8421051, PMID:18591255
- Acts as a negative regulator of VRK1 and VRK2 kinase activity, and enhances AR-mediated transactivation in a polyglutamine length-dependent manner. Related references: PMID:18617507, PMID:10400640
- Subcellular localization includes the nucleus, nuclear envelope, cytoplasm, and melanosomes.
- Post-translationally modified by acetylation, phosphorylation, and isopeptide bond formation; sumoylation and ubiquitination have been detected.
- Widely expressed across human tissues.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from residues 117–216 of human Ran; confirm specificity by peptide competition or knockdown validation.
- For WB, the predicted molecular weight is ~24 kDa; consider using a reducing SDS-PAGE protocol and a blocking buffer optimized for rabbit monoclonal antibodies.
- The antibody is suitable for ELISA; titrate starting from 1:1000 and validate with recombinant Ran protein or cell lysates.
- Cross-reactivity with mouse and rat Ran is reported; verify endogenous expression levels in your model system.
- Store as recommended upon receipt (shipment on ice bags); avoid repeated freeze-thaw cycles.
