Biological Background: RAMP2 Function and Localization
- RAMP2 functions as an accessory protein that interacts with and modulates the activity of G protein-coupled receptors such as calcitonin gene-related peptide type 1 receptor (CALCRL) and calcitonin receptor (CALCR), and is required for the transport of CALCRL to the plasma membrane. Related references: PMID:9620797
- Together with CALCRL, RAMP2 forms a receptor complex for adrenomedullin. Related references: PMID:22102369, PMID:32296767, PMID:9620797
- Together with CALCR, RAMP2 acts as a receptor complex for calcitonin. Related references: PMID:35324283
- Together with CALCR, RAMP2 also acts as a receptor complex for amylin. Related references: PMID:35324283
- RAMP2 localizes predominantly to the cell membrane.
- It is strongly expressed in lung, breast, immune system, and fetal tissues.
- Structural features include a signal peptide, a transmembrane helix, and disulfide bonds; the protein is glycosylated and alternative splicing produces multiple isoforms.
- The unprocessed precursor has a predicted molecular weight of approximately 19.6 kDa, consistent with the mature protein observed at about 20 kDa.
Experimental Guidance and Technical Tips
- For Western blot (WB), validate the specific band at ~20 kDa in RAMP2-expressing tissues such as lung or breast; run appropriate positive and negative controls.
- For immunohistochemistry (IHC-P), consider heat-induced antigen retrieval and test on known positive tissues (e.g., lung) to confirm staining pattern; include an isotype control.
- When using in ELISA, pair with a carefully selected detection antibody and validate performance using recombinant RAMP2 protein or lysates from transfected cells.
- The antibody cross-reacts with mouse and rat RAMP2; therefore, it can be applied in rodent models, but always verify specificity in your experimental system.
