Biological Background: RALBP1 Function and Localization
- RALBP1, also known as RLIP76 or Ral-interacting protein 1, is a ~76 kDa multifunctional protein encoded by the RALBP1 gene (GeneID: 10928).
- Acts as a downstream effector of RALA and RALB, and functions as a GTPase-activating protein (GAP) that inactivates CDC42 and RAC1 by stimulating their intrinsic GTPase activity. Related references: PMID:7673236
- Participates in Ral signaling-dependent regulation of receptor-mediated endocytosis, including ligand-dependent internalization of EGF and insulin receptors. Related references: PMID:10910768, PMID:12775724
- During mitosis, serves as a scaffold for cyclin B-CDK1-mediated phosphorylation of EPSIN/EPN1, inhibiting endocytosis; also facilitates mitochondrial fission by scaffolding DNM1L phosphorylation at mitochondria. Related references: PMID:12775724, PMID:21822277
- Possesses ATP-dependent transport activity for glutathione conjugates and xenobiotics like doxorubicin, potentially contributing to multidrug resistance in cancer cells.
- Subcellularly localizes to multiple compartments including the cell membrane, cytosol, cytoskeleton (spindle pole), nucleus, and mitochondrion.
- Ubiquitously expressed at low levels, with notably strong expression in erythrocytes.
- Post-translationally modified by acetylation and phosphorylation; classified under keywords such as ATP-binding, GTPase activation, and translocase.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 200–300; consider that the epitope may be partially buried in native conformation, potentially requiring denaturation or appropriate antigen retrieval for immunohistochemistry.
- For IHC-P, standard heat-induced epitope retrieval (e.g., citrate buffer pH 6.0) and optimized primary antibody concentration are recommended; validate in species with confirmed cross-reactivity (human, mouse, rat).
- In IF/ICC, permeabilization with 0.1–0.3% Triton X-100 may improve nuclear or mitochondrial antigen accessibility; include appropriate negative controls.
- ELISA applications benefit from titrating the antibody against the immunogen peptide or recombinant RALBP1 protein to establish optimal coating concentration and detection range.
