Biological Background: RALB Function and Localization

  • RALB encodes Ras-related protein Ral-B, a small GTPase that cycles between active GTP-bound and inactive GDP-bound conformations.
  • Functions as a multifunctional GTPase regulating gene expression, cell migration, proliferation, oncogenic transformation, and membrane trafficking. Related references: PMID:10393179 PMID:17875936 PMID:18756269
  • Serves as a GTP sensor for dense core vesicle exocytosis, stabilizing the exocyst complex and targeting it to the leading edge of migrating cells (by similarity).
  • Required for suppression of apoptosis, promoting cell survival through downstream effector interactions. Related references: PMID:17875936
  • During late cytokinesis, recruits the exocyst to the midbody to drive abscission, the final step of cell division. Related references: PMID:18756269
  • Involved in ligand-dependent receptor-mediated endocytosis of EGF and insulin receptors, linking Ral-B to growth factor signaling. Related references: PMID:10393179
  • Localizes primarily at the cell membrane and transiently at the midbody during cytokinesis.
  • Post-translationally modified by prenylation for membrane anchorage and methylation; classified under keywords such as GTP-binding, hydrolase, lipoprotein, and alternative splicing.

Experimental Guidance and Technical Tips

  • For Western blotting, a band of approximately 23 kDa is expected; use whole-cell lysates from human, mouse, or rat cell lines as positive controls.
  • Polyclonal antibodies may exhibit lot-to-lot variability; validate each new lot using a known positive control to ensure consistent performance.
  • For ELISA, optimize coating antigen concentration and detection conditions; the recombinant immunogen can serve as a positive control.
  • Consider confirming specificity by siRNA knockdown or overexpression experiments in relevant cell models.
