Biological Background: RAD54B Function and Localization
- DNA repair and recombination protein RAD54B (RAD54 homolog B, also known as RDH54) is a multifunctional ATPase involved in homologous recombination (HR), a major pathway for repairing DNA double-strand breaks, single-stranded DNA gaps, and stalled replication forks. - During HR, RAD54B acts as a molecular motor, guiding RAD51-coated ssDNA along donor dsDNA to facilitate the homology search and strand exchange. Related references: PMID:11782437 PMID:11884632 PMID:16428451
- After strand exchange, RAD54B dissociates RAD51 from nucleoprotein filaments on dsDNA (By similarity).
- RAD54B can also function as a molecular adapter, promoting MDM2-MDM4 heterodimerization to facilitate ubiquitin-dependent degradation of p53, thereby modulating the cellular decision between cell cycle arrest and progression after DNA damage. Related references: PMID:25384516
