Biological Background: RAD50 Function and Localization
- RAD50 is a core component of the MRN complex (MRE11-RAD50-NBS1), and it plays a central role in DNA double-strand break repair, homologous recombination, telomere integrity, and meiosis.
Related references: PMID:15064416, PMID:21757780, PMID:27889449 - The MRN complex mediates DNA end resection, generating single-stranded DNA critical for homologous recombination, and it facilitates the recruitment of repair factors and activation of ATM/ATR kinases after DNA damage.
Related references: PMID:15064416, PMID:27889449, PMID:28867292, PMID:15790808, PMID:16622404 - The complex possesses single-strand endonuclease and double-strand 3′-5′ exonuclease activities (provided by MRE11), while RAD50 binds and tethers DNA ends to regulate MRE11 activity.
Related references: PMID:11741547, PMID:9590181, PMID:9651580, PMID:12805565, PMID:28134932 - RAD50 contains ATP-binding domains; ATP binding induces a closed conformation that optimally positions DNA ends within the MRE11 active site (by similarity).
- RAD50 is predominantly nuclear, localizing to chromosome telomeres and sites of DNA damage; it also participates in processing R-loops and modulating t-loop formation.
Related references: PMID:10888888, PMID:31537797 - RAD50 is expressed at low levels in most tissues, with elevated expression in testis and fibroblasts.
- RAD50 is subject to acetylation and phosphorylation, and is characterized by ATP-binding, coiled-coil, zinc-binding, and hydrolase motifs.
Experimental Guidance and Technical Tips
- For Western blot analysis, expect a band near 154 kDa. Use an appropriate positive control lysate from human, mouse, or rat cells and include a loading control.
- For IHC-P on paraffin-embedded sections, standard antigen retrieval methods (e.g., heat-induced epitope retrieval) may be considered; pilot experiments are recommended.
- For immunoprecipitation (IP), use standard protocols with pre-clearing steps; the antibody is suitable for pulling down endogenous RAD50 complexes from nuclear extracts.
- For ELISA, the peptide immunogen (N-terminal residues 1-100) may be used to validate antibody titer; cross-reactivity with closely related proteins should be ruled out by sequence alignment.
- Due to its cross-species reactivity, this antibody can be employed for comparative studies in human, mouse, and rat models of DNA repair.
