Biological Background: Rad23A Function and Localization
- RAD23A, also known as UV excision repair protein RAD23 homolog A (HR23A/hHR23A), is a multifunctional protein that participates in histone H4K20 demethylation, DNA repair, ubiquitin-dependent protein degradation, transcriptional regulation, and viral replication. - It specifically demethylates mono-, di- and trimethylated 'Lys-20' of histone H4 (H4K20me1, H4K20me2, H4K20me3) into unmethylated forms, which activates transcription of coding genes and repetitive elements. Related references: PMID:32209475

- For Western blotting, a band around 40 kDa is expected. Optimize loading controls and antibody dilution for your specific lysate.
- In immunofluorescence/ICC, expect nuclear staining consistent with RAD23A localization. Use appropriate fixation and permeabilization protocols.
- For ELISA, consider coating conditions with the synthetic peptide or recombinant protein to assess binding. Perform cross-reactivity tests if applied to non-human/mouse species.
