- RAD18 (also known as RNF73, hHR18, hRAD18) is an E3 ubiquitin-protein ligase belonging to the RING finger protein family. It contains a RING-type zinc finger domain essential for its ubiquitin transferase activity.
- RAD18 plays a critical role in postreplication repair of UV-damaged DNA by mediating gap-filling on the daughter strand during replication of damaged DNA.
- It forms a complex with the E2 ubiquitin-conjugating enzyme UBE2B to catalyze the mono-ubiquitination of DNA-associated proliferating cell nuclear antigen (PCNA) on Lys-164.
- RAD18 possesses single-stranded DNA (ssDNA) binding activity, which may facilitate its recruitment to sites of DNA damage.
