Biological Background: RAD17 Function and Localization
- Cell cycle checkpoint protein RAD17, also known as RF-C/activator 1 homolog, is encoded by the RAD17 gene. Alternative names include R24L, RAD24, HRAD17, and RAD17SP.
- RAD17 is expressed as multiple isoforms; isoform 2 and isoform 3 are the most abundant in non-irradiated cells.
- Essential for sustained cell growth, maintenance of chromosomal stability, and ATR-dependent checkpoint activation upon DNA damage, with weak ATPase activity required for chromatin binding.
Related references: PMID:10208430, PMID:11418864, PMID:11687627 - Participates in the recruitment of the 9-1-1 (RAD1-RAD9-HUS1) complex and RHNO1 onto chromatin, and in CHEK1 activation.
Related references: PMID:21659603 - Involved in homologous recombination by mediating recruitment of the MRN complex to DNA damage sites.
Related references: PMID:24534091 - May also serve as a sensor of DNA replication progression.
Related references: PMID:12578958, PMID:14500819, PMID:15538388 - Predominantly localized to the nucleus and associates with chromosomes.
- Overexpressed in various cancer cell lines and in colon carcinoma (at protein level). Highly expressed in testis, specifically within the germinal epithelium of seminiferous tubules. Weakly expressed in seminomas (testicular tumors).
- Classified as a phosphoprotein involved in cell cycle, DNA damage response, and alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (amino acids 391-670) of human RAD17, which may target epitopes relevant for detecting various isoforms and post-translational modifications.
- For Western blot, a band around 77 kDa is expected; consider validating in cell lines with known RAD17 overexpression, such as cancer-derived lines.
- Due to the polyclonal nature of the antibody, it may recognize multiple RAD17 isoforms and potentially cross-react with related proteins; verify specificity in the relevant sample system.
- For immunoprecipitation, optimize antibody concentration to capture RAD17 and its interacting complexes; confirm recovery by Western blot.
