Biological Background: RACK1 Function and Localization
- RACK1 (Receptor for Activated C Kinase 1), also known as GNB2L1, HLC-7, or PIG21, is a scaffolding protein and an integral component of the 40S ribosomal subunit. It contains multiple WD repeat domains that mediate diverse protein–protein interactions.
- As part of the ribosome, RACK1 is involved in translational repression (PubMed:23636399) and initiates ribosome quality control (RQC) by promoting ubiquitination of 40S subunits during stalled translation (PubMed:28132843). Related references: PMID:23636399 PMID:28132843
- It binds and stabilizes activated protein kinase C (PKC), enhancing PKC-mediated phosphorylation, and may recruit PKC to the ribosome to phosphorylate EIF6, thereby regulating translation.
- RACK1 modulates a wide array of signaling pathways and cellular processes, including cell cycle progression (prolongs G0/G1 phase), apoptosis (promotes BAX oligomerization), integrin signaling, and Wnt pathway inhibition.
- Subcellularly, RACK1 is found in the cytoplasm, cell membrane, nucleus, perinuclear region, dendrites, and phagocytic cups, reflecting its multifunctional roles.
