Biological Background: RACK1 Function and Localization

  • RACK1 (Small ribosomal subunit protein RACK1), also known as GNB2L1, HLC-7, PIG21, acts as a scaffolding protein linking signaling pathways to the translational machinery.
  • As a component of the 40S ribosomal subunit, it plays a role in translational repression.
    Related references: PMID:23636399
  • Participates in ribosome quality control (RQC) by promoting ubiquitination of stalled 40S subunits.
    Related references: PMID:28132843
  • RACK1 binds and stabilizes activated protein kinase C (PKC), enhancing PKC-mediated phosphorylation and potentially recruiting PKC to the ribosome to phosphorylate EIF6.
  • Inhibits SRC kinases and prolongs the G0/G1 phase, thereby regulating cell growth.
  • Localizes to the cell membrane, cytoplasm, nucleus, dendrites, and phagocytic cups, reflecting its multifaceted roles.
  • Implicated in host-pathogen interactions, including enhancing HIV-1 Nef phosphorylation, remodeling ribosomes during poxvirus infection, and facilitating IRES-mediated translation for some RNA viruses.
  • Upregulated in hepatocellular carcinoma and highly expressed in activated hepatic stellate cells, suggesting a role in liver disease.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 220–310 of human RACK1, a region highly conserved across human, mouse, and rat; verify cross-reactivity by sequence alignment or in your sample system.
  • For Western blotting, a band of approximately 35 kDa is expected; consider using a total protein loading control and appropriate molecular weight markers.
  • RACK1 is broadly expressed; however, expression may be elevated in liver cancer cell lines or activated hepatic stellate cells, which could serve as positive controls.
  • Given RACK1's involvement in multiple signaling pathways, consider using specific inhibitors (e.g., PKC inhibitors) to study phosphorylation-dependent interactions.
  • When performing ELISA, coating with the recombinant immunogen or cell lysates may be applicable; validate against a known standard curve.
