Biological Background: RAC2 Function and Localization

  • Official name: Ras-related C3 botulinum toxin substrate 2. Also known as GX, small G protein, and p21-Rac2.
  • Functions as a plasma membrane-associated small GTPase, cycling between an active GTP-bound and inactive GDP-bound state to regulate cellular responses. Related references: PMID:30723080
  • Acts as a regulatory subunit of the phagocyte NADPH oxidase complex, mediating the transfer of electrons from NADPH to O2 to produce superoxide (O2(-)). Related references: PMID:1660188
  • Activates PLCG2, promoting signal transduction pathways involved in calcium mobilization and immune cell activation. Related references: PMID:19394299
  • Participates in diverse biological processes including secretory processes, phagocytosis of apoptotic cells, and epithelial cell polarization.
  • Subcellular localization: Predominantly cytoplasmic.
  • Tissue specificity: Exclusively expressed in hematopoietic cells.
  • Post-translational modifications include acetylation, ADP-ribosylation, glycosylation, prenylation, and methylation, which modulate its activity and localization.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the full-length human RAC2 protein (aa 1-192). It may detect both endogenous and recombinant RAC2; however, validation in the relevant sample system is recommended.
  • For Western blotting, expect a band at approximately 21 kDa. Optimize blocking and antibody dilution for your specific samples and conditions.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider using hematopoietic tissues or cell lines due to the hematopoietic-specific expression of RAC2.
  • As a polyclonal antibody, it may cross-react with closely related RAC family members (e.g., RAC1, RAC3). Verify specificity using knockout controls or isoform-specific assays if necessary.
  • The antibody is suitable for ELISA applications; pair with appropriate capture/detection antibodies for sandwich ELISA format after thorough validation.
