Biological Background: RABGGTB Function and Localization
- RABGGTB encodes the beta subunit of geranylgeranyl transferase type II (GGTase-II), also known as Rab geranylgeranyltransferase subunit beta, a key enzyme in post-translational prenylation of Rab proteins.
- It catalyzes the transfer of a geranylgeranyl moiety from geranylgeranyl diphosphate to both cysteines of Rab proteins bearing C‑terminal sequences -XXCC, -XCXC or -CCXX, such as RAB1A, RAB3A, RAB5A and RAB7A. Related references: PMID:7991565
- RABGGTB also functions as the catalytic subunit of the geranylgeranyl transferase type 3 (GGTase-3) complex, which geranylgeranylates FBXL2 and targets it to cellular membranes, where FBXL2 assembles into the SCF(FBXL2) E3 ubiquitin ligase to mediate degradation of membrane‑anchored proteins.
- The GGTase-3 complex further geranylgeranylates the Golgi v‑SNARE protein YKT6 at Cys‑194, a modification essential for Golgi SNARE complex assembly. Related references: PMID:31209342, PMID:32128853
- Key post‑translational modifications of RABGGTB include acetylation and phosphorylation, and its activity depends on zinc and metal‑ion coordination, consistent with its classification as a prenyltransferase containing repeat motifs.
- The antibody cross-reacts with human, mouse, and rat RABGGTB, reflecting high sequence conservation across these species. The protein has a molecular weight of approximately 37 kDa (calculated mass ~36924 Da).
Experimental Guidance and Technical Tips
- The immunogen used to raise this antibody spans amino acids 1–331, covering the full‑length human RABGGTB protein; therefore, the polyclonal antibody is expected to recognize the native protein in Western blotting (predicted band at ~37 kDa).
- When performing WB, consider using a whole‑cell lysate from human, mouse, or rat cell lines as a positive control, and validate reactivity under reducing and non‑reducing conditions if desired.
- For ELISA, the antibody can be used as a capture or detection reagent, but optimal working dilutions should be determined empirically in the specific assay format.
- Because RABGGTB may be post‑translationally modified (acetylated, phosphorylated), band migration may shift slightly; confirm by mass spectrometry or phosphatase treatment if needed.
