Biological Background: RAB5C / RABL Function and Localization
- Ras-related protein Rab-5C (RAB5C), also known as RABL or L1880, is a small GTPase of the Rab family that acts as a molecular switch in intracellular membrane trafficking.
- The protein cycles between an inactive GDP-bound and an active GTP-bound state, regulating transport vesicle formation, movement, tethering, and fusion with target membranes. Related references: PMID:16086013 PMID:17562788
- RAB5C is specifically involved in early endocytic trafficking and is required for the recruitment of the early endosomal autoantigen EEA1 to early endosomes. Related references: PMID:16086013 PMID:17562788
- It plays an essential role in the endocytosis of epidermal growth factor (EGF) and transferrin and their subsequent trafficking through early endosomes. Related references: PMID:16086013 PMID:17562788
- Subcellularly, RAB5C is localized to the cell membrane, early endosome membrane, and melanosome.
- Post-translational modifications include prenylation, phosphorylation, and glycosylation, which regulate its membrane association and activity.
- RAB5C contains conserved GTP-binding and magnesium-binding sites and exists in multiple isoforms due to alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen encompasses nearly the full-length human RAB5C protein (residues 2–216), providing high specificity for the endogenous target.
- For western blotting, anticipate a band at approximately 23 kDa under standard reducing conditions; optimize blocking and antibody dilution in the relevant cell or tissue lysate.
- In immunofluorescence/ICC and immunoprecipitation assays, validate fixation, permeabilization, and lysis buffer conditions for your specific experimental system.
- The antibody exhibits reported cross-reactivity with mouse and rat orthologs; confirm suitability by testing in the appropriate species-derived samples.
