Biological Background: RAB5C Function and Localization
- RAB5C (Ras-related protein Rab-5C) is a member of the Rab family of small GTPases, which are master regulators of intracellular membrane trafficking. It cycles between an inactive GDP-bound form and an active GTP-bound form that recruits downstream effectors to mediate vesicle formation, movement, tethering, and fusion.
- As a critical component of early endocytic trafficking, RAB5C is required for the recruitment of EEA1 to early endosomes and for the endocytosis and trafficking of epidermal growth factor (EGF) and transferrin through early endosomes. This function is essential for receptor-mediated signaling and nutrient uptake. Related references: PMID:16086013 | PMID:17562788
- Subcellularly, RAB5C is localized to the cell membrane, early endosome membrane, and melanosome, consistent with its role in membrane trafficking and organelle identity.
- The protein undergoes alternative splicing, giving rise to multiple isoforms. Post-translational modifications include prenylation for membrane anchoring, phosphorylation, and glycation; it also binds GTP and magnesium as cofactors.
- RAB5C is a phosphoprotein and a lipoprotein, with a predicted molecular weight of approximately 23 kDa (UniProt). These features are important for its dynamic membrane association and signaling functions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 1-216 of human RAB5C (accession NP_958842.1). Upon validation with the relevant sample system, the antibody may detect full-length and potential splice variants; consider using positive controls such as human cell lines known to express RAB5C.
- For Western blotting, note that the predicted molecular weight is 23 kDa. Design your gel percentage and transfer conditions accordingly, and include a pre-stained molecular weight marker to verify band size.
- In ELISA applications, titrate the antibody against the recombinant protein immunogen or relevant human cell lysates to determine optimal working dilutions.
