Biological Background: RAB5A Function and Localization
- RAB5A (Ras-related protein Rab-5A) is a small GTPase and a key regulator of intracellular membrane trafficking, cycling between inactive GDP-bound and active GTP-bound states to control vesicle formation, movement, and fusion. Related references: PMID:10818110, PMID:14617813, PMID:15378032
- It is essential for the fusion of plasma membranes with early endosomes and for early endocytic trafficking, including EGF and transferrin endocytosis. Related references: PMID:16086013, PMID:17562788
- RAB5A recruits EEA1 and RABEP1/Rabaptin-5 to early endosomes, promoting endocytic membrane fusion. Related references: PMID:16086013, PMID:17562788
- It contributes to filopodia extension and is required for the exosomal release of SDCBP, CD63, PDCD6IP and syndecan. Related references: PMID:14978216, PMID:22660413
- RAB5A localizes to the cell membrane, early endosome membrane, melanosomes, cytoplasmic vesicles, ruffles, and the phagosome membrane.
- It is involved in the recruitment of the FERRY complex to early endosomes, linking them with mRNAs for intracellular distribution. Related references: PMID:37267906
- Post-translational modifications include prenylation, phosphorylation, and glycosylation (by similarity); it binds GTP and magnesium, functioning as a hydrolase.
- RAB5A regulates maturation of apoptotic cell-containing phagosomes, likely downstream of DYN2 and PIK3C3 (by similarity).
Experimental Guidance and Technical Tips
- The immunogen covers the full-length human RAB5A protein (aa 1-215), so the antibody may recognize both GDP- and GTP-bound conformations; consider this when interpreting localization and activity states.
- For Western blot (WB), the expected molecular weight is ~24 kDa; validate the band in your specific sample system and include appropriate controls.
- In IHC-P and IF/ICC, antigen retrieval and cell permeabilization steps are critical; optimize protocols for your target tissue or cell line.
- The antibody is predicted to cross-react with mouse and rat RAB5A, but sequence alignment and experimental validation in these species are recommended.
- For ELISA applications, consider using the recombinant immunogen or purified protein as a positive control.
