Biological Background: RAB3GAP2 Function and Localization
- RAB3GAP2 encodes the non-catalytic 150 kDa subunit (p150) of the Rab3 GTPase-activating protein (Rab3GAP) complex, also known as Rab3-GAP150 or RGAP-iso. The gene undergoes alternative splicing, yielding multiple isoforms.
- As the regulatory subunit of the Rab3GAP complex, RAB3GAP2 accelerates GTP hydrolysis on Rab3 subfamily members (RAB3A, RAB3B, RAB3C, RAB3D), as well as RAB5A and RAB43, thereby acting as a GTPase-activating protein (GAP). Related references: PMID:9733780, PMID:39779760
- The complex also functions as a guanine nucleotide exchange factor (GEF) for RAB18, promoting GDP release and conversion of inactive RAB18-GDP to active RAB18-GTP. Related references: PMID:24891604, PMID:39779760
- RAB3GAP2 mediates the recruitment and stabilization of RAB18 at the cis-Golgi membrane in fibroblasts and at the endoplasmic reticulum (ER) to maintain proper ER architecture. Related references: PMID:26063829, PMID:24891604
- The protein localizes to the cytoplasm and endoplasmic reticulum and is ubiquitously expressed across tissues.
- RAB3GAP2 is essential for normal eye and brain development; it likely participates in neurodevelopmental processes such as cell proliferation, migration, differentiation, and non‑synaptic neurotransmitter release. Mutations are linked to Warburg Micro syndrome (WARBM2), Martsolf syndrome, and spastic paraplegia type 69 (SPG69).
Experimental Guidance and Technical Tips
- The immunogen comprises the N‑terminal 200 amino acids of human RAB3GAP2. Consider this region when assessing potential cross‑reactivity with isoforms or orthologs.
- For Western blot, the predicted full‑length protein migrates at ~156 kDa. Include appropriate positive and negative controls, and validate the antibody in your specific sample matrix.
- The antibody is reported to cross‑react with rat; however, we recommend verifying reactivity in your experimental system using species‑matched tissue or cell lysates.
- For ELISA, optimize coating and detection conditions; as this is a polyclonal antibody, lot‑to‑lot consistency should be confirmed for quantitative applications.
