Biological Background: RAB38 Function and Localization
- Ras-related protein Rab-38, also known as Melanoma antigen NY-MEL-1, belongs to the small GTPase Rab family.
- Small GTPases of the Rab family are key regulators of intracellular membrane trafficking, cycling between an inactive GDP-bound form and an active GTP-bound form that recruits downstream effectors responsible for vesicle formation, movement, tethering, and fusion.
- RAB38 is involved in melanosomal transport and docking, contributing to the proper sorting and peripheral melanosomal distribution of TYRP1 in melanocytes, in cooperation with ANKRD27 and VAMP7.
- A role in phagosome maturation has been reported; RAB38 participates in the maturation of phagosomes that engulf pathogens such as S.aureus and M.tuberculosis. Related references: PMID:21255211
- RAB38 is important for the control of melanin production and melanosome biogenesis, and together with RAB32, regulates the trafficking of melanogenic enzymes TYR, TYRP1, and DCT/TYRP2 to melanosomes. Related references: PMID:23084991
- Subcellular localization includes the cell membrane, melanosomes, cytoplasmic vesicles, and phagosome membranes.
- Predominantly expressed in melanocytes.
- Possesses conserved GTP-binding and hydrolysis domains and undergoes post-translational modifications such as palmitoylation and prenylation, typical of Rab GTPases.
Experimental Guidance and Technical Tips
- The immunogen used corresponds to amino acids 92–211 of human RAB38, a region conserved across human, mouse, and rat, likely contributing to the reported cross-reactivity.
- For Western blot, the predicted molecular weight is approximately 24 kDa; consider validating with positive control lysates from melanocyte or melanoma cell lines.
- For ELISA, test with recombinant RAB38 protein or cell lysates, and optimize coating and detection conditions for your specific experimental system.
- Since RAB38 expression is tissue-specific, use appropriate biological samples and consider enrichment strategies for low-abundance targets.
