Biological Background: RAB31 Function and Localization

  • RAB31, also known as Rab-22B, is a member of the Ras-related Rab subfamily of small GTPases involved in intracellular membrane trafficking regulation. Related references: PMID:11784320
  • It cycles between an inactive GDP-bound form and an active GTP-bound form, recruiting effectors for vesicle formation, movement, tethering, and fusion.
  • RAB31 is essential for Golgi apparatus integrity and normal function, particularly at the trans-Golgi network (TGN).
  • It participates in insulin-stimulated translocation of GLUT4 to the plasma membrane, playing a role in glucose transport.
  • RAB31 mediates transport of mannose-6-phosphate receptor (M6PR) from the TGN to endosomes, and facilitates internalization of EGFR from the cell surface into endosomes.
  • The protein also contributes to phagosome maturation, aiding in the defense against pathogens such as S. aureus and M. tuberculosis.
  • Subcellularly, RAB31 localizes to the Golgi apparatus, trans-Golgi network membrane, early endosomes, and phagosome membranes.
  • Highest tissue expression is observed in placenta and brain, with lower levels in heart and lung; it is undetectable in liver, skeletal muscle, kidney, or pancreas.

Experimental Guidance and Technical Tips

  • The immunogen spans nearly full-length RAB31 (aa 1-195), which may provide robust detection of endogenous RAB31 across various species.
  • For western blot, a band at ~22 kDa is expected; consider optimizing blocking and antibody dilution for your specific sample type.
  • ELISA applications may benefit from using the recombinant immunogen as a positive control or coating antigen.
  • Given cross-reactivity with mouse and rat, the antibody can be used to study RAB31 in rodent models, though validation in relevant tissue lysates is advised.
