Biological Background: Ras-related protein Rab-2A Function and Localization
- RAB2A is a member of the small GTPase Rab family, cycling between active GTP-bound and inactive GDP-bound states to regulate intracellular membrane trafficking.
- It promotes autophagosome-lysosome fusion by recruiting the HOPS endosomal tethering complex, a process involving autophagosomal RAB2A and lysosomal RAB39A. Related references: PMID:37821429
- RAB2A is required for protein transport from the endoplasmic reticulum to the Golgi complex and maintains the compacted morphology of the Golgi. Related references: PMID:26209634
- Together with the closely related RAB2B, RAB2A is redundantly required for efficient autophagic flux. Related references: PMID:28483915
- Subcellular localization includes the endoplasmic reticulum-Golgi intermediate compartment, melanosome, ER membrane, Golgi apparatus membrane, cytoplasmic vesicles, secretory vesicles, acrosome, and autophagosome membrane.
- Post-translational modifications include prenylation for membrane anchoring and acetylation, as indicated by UniProt keywords.
- The protein belongs to the small GTPase superfamily, possessing GTP-binding and hydrolase activities, and is involved in key transport and trafficking pathways.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-212, covering the full-length human RAB2A; it may recognize both native and denatured protein forms.
- For Western blotting, consider optimizing the antibody dilution based on your specific sample and detection system; a band near 24kDa is expected.
- In ELISA applications, validate coating conditions and detection system using a positive control (e.g., recombinant RAB2A protein).
- Verify antibody specificity in biologically relevant samples from human, mouse, or rat due to reported cross-reactivity.
- Because this is a polyclonal antibody, lot-to-lot consistency may vary; include positive and negative controls in each experiment.
